What is the principle of incubator shaker?
An incubator shaker works by combining controlled temperature incubation with continuous shaking motion to create an optimal environment for the growth and cultivation of microorganisms, cells, and biological samples.
The principle is based on two core functions:
Temperature control
Agitation (shaking)
working together to promote uniform mixing, oxygen transfer, and stable culture conditions.
How to use a shaking incubator?
- Prepare the Samples
Before operation:
Place culture media or samples into suitable flasks or containers
Ensure caps are loosened if gas exchange is required
Check that flasks are properly sealed to avoid leakage - Load the Platform Correctly
Secure the containers using:
Flask clamps
Tube racks
Universal platforms
Distribute the load evenly across the shaking platform to maintain balance and reduce vibration. - Set the Temperature
Adjust the incubation temperature according to experimental requirements.
Typical settings include:
37℃ for bacterial culture or lower temperatures for protein expression studies.
Allow the chamber to preheat before starting sensitive experiments. - Set the Shaking Speed
Adjust the shaking speed (rpm) depending on the sample type and oxygen demand.
Typical shaking ranges are:
2–300 rpm
Higher speeds generally improve oxygen transfer but may increase foaming or shear stress. - Set the Operating Time
Use the timer function if required.
Some incubator shakers support:
Continuous operation
Timed operation
Programmable multi-step protocols - Start the Incubator Shaker
After confirming:
Temperature settings
Speed settings
Secure sample placement close the chamber door and start operation.
Monitor the system during the first few minutes to ensure stable shaking. - Monitor During Operation
During incubation:
Check temperature stability
Observe abnormal noise or vibration
Ensure samples remain secured
Avoid opening the door frequently, as this may affect temperature consistency. - Remove Samples Safely
After the experiment:
Stop shaking first
Open the chamber carefully
Remove samples slowly to avoid spills
Use appropriate PPE if handling biological or hazardous materials. - Clean the Equipment
After use:
Wipe the chamber and platform
Remove spills immediately
Clean clamps and accessories
Disinfect if biological samples were used
Routine cleaning helps prevent contamination and extends equipment lifespan.
